大孔树脂纯化川芎中阿魏酸的工艺优选 被引量:1

Purification Optimization of Ferulic Acid in Ligusticum Chuanxiong Hort. by Macroporous Resin

摘要 目的:优选大孔树脂纯化川芎中阿魏酸的工艺条件。方法:提取液预处理后,用D101、HPD100、AB-8型大孔树脂纯化其阿魏酸,通过静态吸附、解吸试验优选树脂型号,考察上样量、上样液质量浓度、pH、吸附流速、洗脱剂种类及用量等工艺参数。结果:D101型大孔树脂对阿魏酸的吸附率最好,解吸率最高,最佳富集工艺为上样量8 BV,上样液质量浓度0. 25 mg·ml^(-1),pH为5,上样流速3 BV·h^(-1),用5 BV水洗,再用50%乙醇6 BV洗脱,洗脱流速5 BV·h^(-1)。阿魏酸转移率为50. 36%,出膏率为2. 52%。结论:该工艺稳定可行,可用于川芎中阿魏酸的纯化。 Objective:To optimize the purification conditions of ferulic acid in Ligusticum chuanxiong Hort by macroporous resin.Methods:After the pretreatment extract,ferulic acid was purified by D101,HPD100 and AB-8 macroporous resin.The resin type was optimized by static adsorption and desorption test.Sample solution volume,sample concentration,sample pH,sample flow rate,eluent type,eluent amount and the other parameters were investigated.Results:D101 type macroporous resin had the best adsorption and desorption capacity for ferulic acid with following technology conditions:the sample volume of 8 BV,the sample concentration of 0.25 mg·ml^-1,the sample pH of 5 and the sample flow rate of 3 BV·h^-1,washed by 5 BV of water,eluted by 6 BV of 50%ethanol,and eluting velocity of 5 BV·h^-1.The transfer rate of ferulic acid was 50.36%,and the paste-forming rate was 2.52%.Conclusion:The process is stable and feasible,and can be used for the purification of ferulic acid in Ligusticum chuanxiong Hort.
