乳移平对乳腺癌细胞增殖和迁移能力影响及机制研究

Effects of Ruyiping on proliferation and migration of breast cancer cells

目的研究中药乳移平对MDA-MB-231细胞增殖与侵袭能力的影响及其分子机制。方法使用乳移平刺激MDAMB-231细胞,通过MTT实验检测细胞增殖,通过Transwell实验检测细胞侵袭能力。RT-PCR法和免疫印迹法检测乳移平刺激后及加入NF-κB抑制剂后E-Cadherin表达水平。结果乳移平对MDA-MB-231细胞增殖能力有一定抑制能力,并且其能显著抑制细胞迁移;乳移平能够通过NF-κB通路诱导E-Cadherin表达来抑制乳腺癌细胞迁移。结论乳移平能够抑制乳腺癌细胞MDA-MB-231细胞增殖和迁移能力,其机制可能与促进E-Cadherin表达有关。

Objective To study the effects of Ruyiping on the proliferation and invasion of MDA-MB-231 cells and its mechanism.Methods MDAMB-231 cells were treated with Ruyiping.The proliferation ability was detected by MTT and the invasion ability was detected by Transwell experiment.The expression of E-Cadherin was also detected by real-time PCR and Western blot in MDA-MB-231 cells after Ruyiping and NF-κB inhibitor stimulation.Results Ruyiping slightly inhibited MDA-MB-231 proliferation and significantly suppressed the migration of tumor cells by inducing ECadherin expression through NF-κB pathway.Conclusion Ruyiping can inhibit the proliferation and migration of MDA-MB-231 cells and its mechanism may be related to the promotion of E-Cadherin expression.
