旱半夏组培苗生根条件的优化 被引量:8

Optimization of Rooting Condition of Tissue Cultured Seedling of Pinellia ternata

摘要 本研究以野生旱半夏的组培苗为材料,对NAA和IBA两种激素浓度,培养基中无机盐的浓度及活性炭的浓度对组培苗生根的影响进行研究,选用了平均生根数、平均根长、生根率、根的粗细质量及移栽成活率等指标评价不同条件对旱半夏组培苗生根的影响。结果显示,低浓度的NAA利于生根,IBA处理的整体效果较好,较低浓度的无机盐促进根的生长,活性炭的加入能较明显的促进组培苗根的伸长,使根更粗壮。因此,旱半夏组培苗在1/2 MS+0.05 mg/L NAA+0.5 g/L AC最合适生根。本研究优化了旱半夏组培苗的生根条件,为旱半夏组培苗的移栽及其生物技术育种推广提供基础支持。 In this study,using tissue cultured seedling of wild Pinellia ternata as material,the effects of the concentrations of two hormones,NAA and IBA,the concentration of inorganic salts in the medium and the concentration of activated carbon on the rooting of tissue cultured seedling were studied.The effects of different conditions on the rooting of tissue cultured seedlings were evaluated by using such indicators as average number of roots,average root length,rooting rate,thickness and quality as well as survival rate of roots.The results showed that low concentration of NAA was beneficial to rooting,and the overall effect of IBA treatment was better.Lower concentration of inorganic salt promoted the growth of roots and the addition of activated carbon could obviously promote the elongation of the tissue cultured seedling roots,as well as make the roots stronger.Therefore,it was optimum for the seedlings to take root in 1/2 MS+0.05 mg/L NAA+0.5 g/L AC.The optimized rooting conditions of the tissue cultured seedling of Pinellia ternata would provide basic support for the transplanting and the popularization ofbiotechnological breeding.
