摘要
本研究用60%甲醇和60%乙醇提取杜仲愈伤组织与杜仲叶和杜仲皮的活性成分,测定其总黄酮和总酚含量,比较分析其抗氧化活性及降血糖能力。结果表明,杜仲叶中总黄酮含量和总酚含量最高,杜仲愈伤组织次之,杜仲皮最低。杜仲叶中60%甲醇和60%乙醇提取物中总黄酮含量分别达到283.738 mg QuE/g DM和285.065 mg Qu E/g DM,总酚含量分别达到59.100 mg GAE/g DM和66.720 mg GAE/g DM。抗氧化活性分析表明,60%甲醇提取物DPPH·和ABTS+·清除能力均为叶>愈伤组织>皮,对Fe2+络合能力为愈伤组织>叶>皮。60%乙醇提取物对DPPH·和ABTS+·清除能力均为叶>皮>愈伤组织,对Fe2+络合能力为叶>愈伤组织>皮。60%甲醇和60%乙醇提取物对α-葡萄糖苷酶活性抑制能力均为叶>皮>愈伤组织。相关性分析表明,杜仲总黄酮、总酚与DPPH·清除能力的相关系数分别为-0.975、-0.972,与ABTS+·清除能力的相关系数分别为-0.988、-0.986,表明总黄酮和总酚与DPPH·清除能力、ABTS+·清除能力具有显著相关性。总黄酮、总酚与Fe2+络合能力的相关系数分别为-0.541、-0.514,说明总黄酮、多酚与Fe2+络合能力之间有一定的相关性。总黄酮、总酚与α-葡萄糖苷酶活性抑制能力的相关系数分别为-0.871、-0.883,说明总黄酮、多酚与α-葡萄糖苷酶活性抑制能力具有显著相关性。本研究为传统药用部位杜仲皮的替代品开发和利用提供科学依据。
Active ingredients ofEucommia ulmoides callus,leaf and bark were extracted with 60%methanol and 60%ethanol in this paper.The content of total flavonoids and total phenolic of the extracts were determined,and the antioxidant activity and hypoglycemic ability were compared.The results showed that the content of total flavonoids and total phenolic of E.ulmoides leaves were the highest,followed by that of callus,and that in bark was the lowest.The total flavonoids content of extract from E.ulrnoides leaves extracted with 60%ethanol and 60%ethanol reached 283.738 mg QuE/g DM and 285.065 mg QuE/g DM,and the total phenol content reached 59.100 mg GAE/g DM and 66.720 mg GAE/g DM,respectively.The antioxidant activity analysis showed that the ability of 60%methanol extract to scavengeDPPH·and ABTS^+·was leaf >callus >bark.The scavenging ability of 60%ethanol extracts to DPPH·and ABTS^+·was leaf >bark >callus,while the complexing power to Fe^2+ was leaf >callus >bark.The inhibitory ability of 60%methanol and 60%ethanol extracts to the activity of alpha glucosidase both was leaf >bark >callus.The correlation analysis indicated that the correlation coefficient of total flavonoids and total phenolics content of E.ulmoides extract with DPPH ~and ABTS+~scavenging ability were -0.975,-0.972,and -0.988,-0.986,respectively,indicating that the total fiavonoids and total phenolic content was significantly correlated to DPPH ~scavenging ability and ABTS+.scavenging ability.The correlation coefficient of total flavonoids and total phenolics content with Fe2+complexing power was -0.541 and -0.514,implying that there was a certain correlation between total flavonoids and total phenolics content and Fe2+complexing power. The correlation coefficient of total flavonoids content and total phenols content with activity of alpha glucosidase inhibitory capacity were -0.871,-0.883,indicating that the tota!flavonoids content,polyphenols content and alpha glucosidase inhibitory capacity were significantly correlated.This paper could provide a scientific basis for the development and utilization of the replacement products of the traditional medicinal parts orE.ulmoides bark.
