摘要
[目的]探讨牛膝多糖(ABP)对四氯化碳(CCl_4)所诱导的大鼠急性肝损伤的保护作用.[方法]取Wistar大鼠,随机分为正常对照组、模型对照组、ABP 100,200 mg/kg组、水飞蓟素组,分别灌胃给予生理盐水、ABP和水飞蓟素7 d,最后1 d腹腔注射给予CCl_4,16 h后检测血清中ALT,AST,ALP水平;行HE染色观察肝脏病理学变化;利用黄嘌呤氧化酶法和硫代巴比妥酸法分别检测肝匀浆中SOD,CAT,MDA水平;Western blot法检测肝组织中NF-κB p65蛋白的表达.[结果]与正常对照组比较,模型对照组大鼠血清中ALT,AST,ALP水平显著升高(P<0.05),肝组织变性坏死累及全小叶.与模型对照组比较,ABP 100,200 mg/kg组血清中ALT,AST,ALP水平显著降低(P<0.05),肝组织病变程度较轻,胞浆中SOD,CAT活性显著升高(P<0.05),MDA水平显著降低(P<0.05).模型对照组肝组织中NF-κB p65蛋白表达显著高于正常对照组(P<0.01),而ABP 100,200 mg/kg组NF-κB p65蛋白表达较模型对照组显著降低(P<0.01),且呈剂量依赖性.[结论] ABP对CCl_4所诱导的大鼠急性肝损伤具有保护作用,其机制认为可能与ABP抗脂质过氧化和抗炎症反应有关系.
OBJECTIVE To investigate the protective effects of achyranthes bidentata polysaccharides(ABP) on acute liver injury induced by carbon tetrachloride(CCl4) in rats. METHODS Wistar rats were randomly divided into normal control group, model control group, ABP 100, 200 mg/kg groups and silibinin group, they were given normal saline, ABP and silibinin by gavage for 7 d.On the last day, they were administered with CCl4 by intraperitoneal injection. The levels of ALT, AST and ALP in serum were detected. Pathological changes of hepatic tissue were observed by HE staining. The levels of SOD, CAT and MDA in liver homogenate were detected by xanthinoxidase method and thiobarbituric acid method. Western blot was used to detect the expression of NF-κB p65 protein in the liver. RESULTS Compared with the normal control group, the serum levels of ALT,AST and ALP in the model control group were significantly increased(P<0.05), and the degeneration and necrosis of liver tissue involved the entire lobule. Compared with the model control group, the serum levels of ALT, AST and ALP in the ABP 100,200 mg/kg groups were significantly decreased(P<0.05), the lesion degree of liver tissue was mild,and the levels of SOD and CAT in liver homogenate were increased(P<0.05), while the level of MDA was significantly decrease(P<0.05). The expression of NF-κB p65 protein in the liver of the model control group was obviously higher than that in the normal control group(P<0.01),while its expression in the liver of the ABP 100,200 mg/kg groups was significantly lower than that in the model control group(P<0.01), and the results were dose dependent. CONCLUSION ABP had protective effects on the acute liver injury induced by CCl 4, and its mechanism might be related with the anti-lipid peroxidation and anti-inflamation reaction of ABP.
