欧前胡素对哮喘气道慢性炎症及气道重塑的抑制作用 被引量:4

摘要 [目的]探讨欧前胡素对哮喘模型小鼠气道炎症及气道重塑的抑制作用.[方法]取40只雄性BALB/C小鼠,随机分为5个组,利用卵清蛋白(OVA)诱导制作小鼠哮喘气道重塑模型,分别给予10,30mg/kg欧前胡素及0.5mg/kg地塞米松治疗.在最后1次激发结束24h后处死小鼠,取肺泡灌洗液(BALF)及肺组织备用.取肺组织行包埋切片后行HE,PAS,Masson染色,观察肺组织病理学变化,行Diff-Quick染色对BALF中的细胞进行分类和计数,利用ELISA法检测BALF中IL-4,IL-5,IL-13和IFN-γ含量,利用Western blot法对肺组织中的!-SMA表达水平进行定量分析.[结果]欧前胡素可明显抑制哮喘模型小鼠肺组织中炎性细胞浸润、杯状细胞增生,减少黏液分泌及胶原沉积;亦可减少BALF中炎症细胞数量,降低BALF中IL-4,IL-5,IL-13水平,升高IFN-γ水平,抑制!-SMA表达水平,与OVA哮喘组比较差异均有统计学意义(P<0.05).[结论]欧前胡素可抑制哮喘模型小鼠气道炎症反应及气道重塑. OBJECTIVE To investigate the inhibitory effects of imperatorin on airway inflammation and airway remodeling in asthmatic mice.METHODS40 male BALB/C mice were selected and randomly divided into five groups.The model of mouse asthmatic airway remodeling was induced by ovalbumin(OVA).They were treated with 10 and 30 mg/kg imperatorin and 0.5 mg/kg dexamethasone respectively.The mice were sacrificed 24 hours after the last stimulation,and the bronchoalveolar lavage fluid(BALF)and lung tissues of the mice were obtained for later use.The lung tissues were embedded and HE,PAS and Masson staining were performed to observe the histopathological changes of lung tissue.Diff-Quick staining was used to sort and count the cells in the BALF.ELISA was used to detect the content of IL-4,IL-5,IL-13 and IFN-γin the BALF.Western blot was used to analyze the expression of!-SMA in lung tissue.RESULTSImperatorin could significantly inhibit the inflammatory cell infiltration,goblet cell proliferation,mucus secretion and collagen deposition in the lung tissues of asthma model mice.It could also reduce the number of inflammatory cells and decrease the levels of IL-4,IL-5 and IL-13,and increase the content of IFN-γin the BALF.It could inhibit the expression of!-SMA.The difference was statistically significant as compared with the OVA group(P<0.05).CONCLUSIONImperatorin could inhibit airway inflammation and airway remodeling in asthma model mice.
