摘要
[目的]分析延边地区朝鲜族和汉族小学生不同代谢指标与一氧化氮合酶3(NOS3)基因单核苷酸多态性(SNPs)的关系.[方法]选择2011年8月—2013年8月间吉林省延边朝鲜族自治州3所小学校的1 206名小学生(朝鲜族706名,汉族500名)作为调查对象,测量体质量指数(BMI)、腰围(WC)、收缩压(SBP)和舒张压(DBP)值;检测空腹血清三酰甘油(TG)、高密度脂蛋白胆固醇(HDL-C)、血糖(FBG)、胰岛素(FIns)值和NOS3基因rs3918227位点的基因型.[结果]朝鲜族小学生NOS3基因SNP rs3918227位点CC,CA及AA型的频率分别为93.5%,6.2%和0.3%,汉族小学生分别为84.4%,15.6%和0.0%,朝鲜族和汉族小学生的基因型分布频率间差异有统计学意义(P<0.001);朝鲜族小学生C,A等位基因频率分别为96.6%和3.4%,汉族小学生分别为84.4%和15.6%,朝鲜族和汉族小学生的等位基因频率分布间差异有统计学意义(P<0.001).NOS3基因SNP rs3918227位点不同基因型朝鲜族小学生WC,SBP,DBP和HOMA-IR分布特征间均存在显著差异(P<0.05),汉族小学生BMI和FBG分布特征间均存在显著差异(P<0.01).[结论]延边地区朝鲜族和汉族小学生NOS3基因rs3918227位点具有SNPs,且2个民族频率分布间存在差异;朝鲜族和汉族小学生多种代谢指标与NOS3基因rs3918227位点基因型和等位基因的关联程度有所不同.
OBJECTIVE To analyze the relationship between the different metabolic indexes and single nucleotide polymorphism(SNPs)of nitric oxide synthase 3(NOS3)gene in Korean and Han nationality pupils of Yanbian area.METHODS 1 206 pupils(706 Korean nationality and 500 Han nationality)from 3 primary schools in Yanbian Korean autonomous prefecture were selected from August 2011 to August 2013.The body mass index(BMI),waist circumference(WC),systolic blood pressure(SBP)and diastolic blood pressure(DBP)were measured.Blood triglyceride(TG),high-density lipoprotein cholesterol(HDL-C),fasting blood glucose(FBG),insulin(FIns)and genotypes of NOS3 gene rs3918227 site were measured after 12 hour fasting.RESULTS The frequencies of CC,CA and AA genotype at the SNP rs3918227 site of NOS3 gene were 93.5%,6.2% and 0.3% in the Korean nationality pupils and 84.4%,15.6%and 0.0%in the Han nationality pupils, and there was statistically significant difference in genotype frequencies between Korean and Han nationality pupils(P<0.001).The frequencies of C and A allele were 96.6% and 3.4% in Korean nationality pupils and 84.4% and 15.6% respectively in Han nationality pupils,and there was significant difference in the distribution of allele frequencies between them(P<0.001).There were significant differences among the distribution characteristics of WC,SBP,DBP and HOMA-IR in Korean nationality pupils with the different genotype of SNP rs3918227 site of NOS3 gene(P<0.05),and there were different distribution characteristics of BMI and FBG in Han nationality pupils(P<0.01).CONCLUSION NOS3 gene rs3918227 site had SNPs in the Korean and Han nationality pupils of Yanbian area,and there were differences in frequency distribution between the two nationalities.The degree of association between various metabolic indexes and genotypes and alleles of NOS3 gene rs3918227 site was different in Korean and Han nationality pupils.
