摘要
Sirtuins(SIRT)是一类具有ADP-核糖基转移酶和(或)脱乙酰酶活性的蛋白质,在哺乳动物中有7名家族成员(SIRT1~7),广泛分布于心肌细胞、血管平滑肌细胞、内皮细胞等心血管与肾脏系统中。SIRT参与调节心血管重构、氧化应激、炎症免疫、凋亡自噬及衰老等病理生理过程,其活性或表达异常参与冠心病、高血压、慢性心力衰竭等心血管疾病的发生发展。本文主要介绍SIRT家族结构功能及其与心血管疾病研究新进展。
Sirtuins(SIRT) are proteins with NAD+-dependent deacetylases and/or ADP-ribosyl transferases, which have seven members from SIRT1 to SIRT7 in mammalian and are widely distributed in cardiomyocytes, vascular smooth muscle cells, endothelial cells in cardiovascular system and kidney system. SIRT play an important role in the regulation of cardiovascular remodeling, oxidative stress, inflammation and immune reaction, apoptosis, autophagy, senescence and other pathophysiological processes. Their abnormalities in activities or expression are implicated in occurrence and development of cardiovascular diseases, such as coronary heart disease, hypertension and chronic heart failure. In this paper, we will give a brief introduction of the structure and functions of SIRT family and the research progress of SIRT signaling in cardiovascular diseases.
