我国家养雉鸡MHC B-F基因表达量测定与分析 被引量:1

Determination and Analysis of MHC B-F Gene Expression of Domestic Pheasants in China

摘要 主要组织相容性复合体(Major histocompatibility complex,MHC)是脊椎动物基因组中最具多态性和基因密集的区域。MHC基因在免疫系统中发挥着重要的作用,能够参与机体内、外抗原的呈递,从而影响机体对疾病的抵抗力。鸡是除哺乳类动物外,最早及最深入研究MHC基因的一类脊椎动物,研究证实MHC基因是鸡抗病育种研究中的优选标记基因。试验利用罗氏实时荧光定量PCR技术对我国家养的5个雉鸡种群(中国环颈雉、白雉鸡、日本绿雉、申红雉鸡和黑化雉鸡) MHC B-F基因在不同组织中表达含量进行了测定和比较分析。结果表明:MHC B-F基因在各种群雉鸡的7个组织中均有表达,其中肝脏中MHC B-F基因相对表达量最高,肌胃中相对表达量最少。在所测的7个组织中除腺胃外,申红雉鸡MHC B-F基因的相对表达量普遍高于其他种群。试验研究了MHC B-F基因在我国家养雉鸡不同组织器官中的表达量差异,为进一步研究MHC B-F基因对抗感染性疾病的影响及其对疾病的抵抗力提供了基础数据。 The major histocompatibility complex(MHC) is the most polymorphic and gene intensive region of vertebrate genome. The MHC gene plays an important role in the immune system and is involved in the presentation of both internal and external antigens, thereby affecting the body’s resistance to disease. So far, chickens have been the first and most intensively studied MHC genes except mammals. Studies have confirmed that MHC gene is the preferred marker gene in the study of chicken resistance breeding. This experiment uses Roche real-time fluorescence quantitative PCR in five populations of our domestic pheasant(Chinese ring pheasant, white pheasant, Japanese green pheasant, Shenhong pheasant and black pheasant) to measure and analyze MHC B-F gene expression levels in different tissues. The results show that MHC B-F gene is expressed in seven tissues of all groups of pheasant. The relative expression of MHC B-F gene in liver is the highest and the relative expression in stomach is the least. The relative expression of the MHC B-F gene in Shenhong pheasant is generally higher than that of other populations in the seven tissues surveyed. This experiment studies MHC B-F gene expression quantity differences in different tissues and organs of China′s domestic pheasants, which provides basic data for studying the effect of MHC B-F gene on fighting against infectious disease and resisting disease.
