鞘膜内应用鼠神经生长因子及甲泼尼龙治疗大鼠坐骨神经离断伤的实验研究

摘要 目的:研究鞘膜内应用鼠神经生长因子(mouse nerve growth factor mNGF)及甲泼尼龙治疗大鼠坐骨神经损伤的疗效。方法:将30只健康成年的wistar大鼠按随机数字分组法分为A、B、C3组,每组10只,将大鼠坐骨神经切断后再吻合。A组鞘膜内注射鼠神经生长因子与甲泼尼龙(mNCF 10μg/kg与甲泼尼龙10mg生理盐水稀释成0.1ml液体),B组鞘膜内注射甲泼尼龙(甲泼尼龙10mg生理盐水配成0.1ml液体),C组鞘膜内注射0.1ml生理盐水。实验结束后,观察动物大体形态,电生理方面分析比较损伤神经传导速度,形态学标志中分析比较镜下切片中有髓轴突数目和髓鞘厚度情况。结果:损伤神经传导速度比较,A组快于B、C2组;镜下切片中有髓轴突数目和髓鞘厚度情况比较,A组均大于B、C2组;以上差别具有统计学意义(P<0.05)。结论:鞘膜内应用mNGF及甲泼尼龙可有效地促进大鼠离断坐骨神经吻合后的功能恢复。 Objective:To analyses the curative effect of mouse nerve growth factor (mNGF)and methylprednisolone intrathecal ad- ministration on rat sciatic nerve mutilation injury.Methods:Thirty healthy and ripe Wistar rats were randomly divided into 3 groups(n = 10 each),Suturing the rat sciatic nerve after cut -off,The group A received intrathecal injection of mNGF and methylprednisolone (mNGF 10μg/Kg,methylprednisolone 10mg and isotonic Na chloride attenuated to 0.1ml),while group B was given methylprednisolone only(methylprednisolone 10mg and isotonic Na chloride attenuated to 0.lml),and the group C received intrathecal injection of isotonic Na chloride (0.1 ml ).To analysis and compare neural stem conduction velocity relating to electrophysiology signal,myelinated nerve fiber count and thickness of myelin sheath relating to morphology signal at the end of experimentation.Results:Intrathecal application of mNGF and methylprednisolone group were superior to the other two groups with significant difference (P<0.05)in neural stem conduction velocity,myelinated nerve fiber count,thickness of myelin sheath.Conclusion:The treatment of intrathecal application of mNGF and methylprednisolone can effectivly promote repair and regeneration after inosculation of rat sciatic nerve mutilation injury.
