微小核糖核酸-21-3p在小鼠胰腺炎模型中的表达和意义 被引量:6

Expression and significance of microRNA-21-3p in mouse pancreatitis model

摘要 目的 探讨微小核糖核酸(miRNA)-21-3p在小鼠急性胰腺炎(AP)模型中的表达和意义。方法 左旋-精氨酸注射构建小鼠胰腺炎模型,以随机数表法随机分为5组:对照组,急性胰腺炎模型组(AP 6 h组、AP 12 h组、AP 24 h组、AP 48 h组),每组10只;再以不同剂量的左旋-精氨酸处理小鼠,分为AP-A、AP-B、AP-C组,收集小鼠胰腺组织及血清,实时定量聚合酶链式反应(RT-PCR)检测小鼠胰腺及血清中miR-21-3p相对表达;酶联免疫吸附实验(ELISA)检测血清淀粉酶、胰腺髓过氧化物酶(MPO),炎性因子肿瘤坏死因子-α(TNF-α)、白细胞介素(IL)-6、IL-17、IL-23表达,Pearson检验验证其相关性。结果 与对照组比较,AP组小鼠胰腺组织及血清中miR-21-3p表达明显升高(P<0.01),miR-21-3p在AP 6、12、24、48 h组相对表达随时间增加(P<0.01),24~48 h达到峰值(P>0.05);AP-A、AP-B、AP-C组miR-21-3p表达较对照组均明显升高(P<0.01),组间miR-21-3p表达也依次升高(P<0.01);AP组小鼠血清淀粉酶、MPO及炎性因子TNF-α、IL-6、IL-17、IL-23表达较对照组明显升高(P<0.01),AP发生6~24 h,miR-21-3p表达与血清淀粉酶、MPO呈正相关(r=0.632、0.601,P<0.01);miR-21-3p表达与TNF-α以及IL-23血清水平呈正相关(r=0.593、0.611,P<0.05)。结论 监测血清miR-21-3p有助于急性胰腺炎的早期诊断和病情评估。 Objective To explore the expression and significance of microRNA-21-3p in mouse acute pancreatitis model.Methods C57BL/6 mouse pancreatitis model was constructed by L-arginine injection and randomly divided into 5 groups: control group, acute pancreatitis model group (AP 6, 12, 24, 48 h group). Then 30 mice were treated with different doses of L-arginine, divided into AP-A, AP-B, and AP-C groups. Mouse pancreatic tissue and serum were collected and real-time quantitative polymerase chain reaction was used to detect the relative expression of miR-21-3p. Enzyme-linked immunosorbent assay (ELISA) was used to detect serum amylase, pancreatic myeloperoxidase (MPO) and inflammatory factor tumor necrosis factor-α (TNF-α), interleukin (IL)-6, IL-17, IL-23 expression, Pearson test verified the correlation between miR-21-3p and serum indicators.Results The expression of miR-21-3p in pancreatic tissue and serum of AP group was significantly increased (P<0.01). The relative expression of miR-21-3p was increased with time (P<0.01), peaking at 24-48 h (P>0.05). In AP-A, AP-B, AP-C group, the expression of miR-21-3p was significantly higher than that of the control group (P<0.01), and increased sequentially (P<0.01). The expressions of serum amylase, MPO and inflammatory factors TNF-α, IL-6, IL-17 and IL-23 in AP group were significantly higher than those in the control group (P<0.01). After AP occurred 6-24 h, miR-21-3p expression was positively correlated with serum amylase and MPO (r=0.632, 0.601, P<0.01). Also, miR-21-3p expression was positively correlated with serum levels of TNF-α and IL-23 (r=0.593, 0.611, P<0.05).Conclusion Monitoring serum miR-21-3p can provide a basis for the early diagnosis and assessment of acute pancreatitis.
