摘要
目的 :研究枸杞多糖 (lyciumbarbarumpolysaccharides ,LBP)对癌细胞周期及凋亡的影响。方法 :用加入不同浓度LBP的PRMI - 16 4 0培养基进行PG细胞培养 ,相差显微镜观察PG细胞形态改变及细胞增殖状况 ,流式细胞仪溴化乙锭 (PI)染色法测定PG细胞DNA含量及凋亡细胞比例。结果 :1)小剂量 (5 0~ 2 0 0 μg/ml)LBP促进PG细胞增殖 ,大剂量 (40 0 μg/ml)LBP则抑制PG细胞增值 ;2 )LBP诱导PG细胞凋亡并存在时间和剂量依赖性。结论 :LBP对PG细胞增殖具有双向调节作用 ;LBP可诱导PG细胞凋亡。
Objective: To study the effect of lycium barbarum polysaccharides (LBP) on the proliferation and apoptosis of cancer cell. Methods: PG cell was cultured in RPMI-1640 medium supplemented with 10% FCS and with various concentration of LBP in 37℃ incubator provided continuously with 5% CO 2. The effects of LBP on PG cellular proliferation and apoptosis were analysed by flow cytometer(Beckton-Dickinson) and the morphological changes of PG cell caused by LBP were observed with phase contrast microscope. Results: 1)50-200μg/ml LBP caused PG cell proliferation, but 400μg/ml LBP inhibited PG cell proliferation. 2)LBP caused apoptosis of PG cell and the induced apoptosis of PG cell by LBP was dose-and-time-dependent. Conclusion:1.LBP shows bidirectional control over PG cell proliferation. 2. LBP induces PG cell apoptosis.
