人胰腺癌细胞FHIT基因转染对肿瘤坏死因子敏感性的影响 被引量:4

摘要 目的 将FHIT基因导入人胰腺癌 1 990细胞并观察其对TNF敏感性的改变 ,探讨FHIT对胰腺癌肿瘤坏死因子 (TNF)敏感性的关系。方法 通过脂质体转染法将外源FHIT导入有FHIT全基因丢失的人胰腺癌 1 990细胞 ,筛选阳性克隆 ,同时以空载体pRC/CMV作为对照 ,加入TNF后作生长曲线并计算细胞成活数。结果 转染FHIT的 1 990细胞阳性克隆与亲代相比 ,其生长速度分别为 32× 1 0 4 cells/well、78× 1 0 4 cells/well(P <0 .0 1 ) ,诱导了 1 990细胞的凋亡 ,且在TNF为 1 0 3μg/L时其细胞存活率分别为 2 3 %、74% (P<0 .0 5)。结论 FHIT基因转染增强了人胰腺癌 1 990细胞对TNF的敏感性 。 Objective In order to determine the effect of wide type FHIT gene on tumor necrosis factor (TNF) sensitivity of human pancreatic adenocarcinoma,wide type FHIT gene was transduced into human pancreatic adenocarcinoma 1990 cells in which FHIT gene was lost totally.Methods By the method of liposome transfection,FHIT gene was transduced into human 1990 cell lines,and the integration and expression of FHIT were detected by RT PCR and western blotting.Then the growth was observed and the cell survival number was acquired after the treatment of TNF.Results Expression of FHIT was detected by RT PCR and western blotting.The growth rate of FHIT transfected 1990 cells and 1990 cells was 32×10 4 cells/well and 78×10 4 cells/well,respectively (P<0.01) with their survival rate being 23% and 74% respectively in the presence of TNF (10 3?μg/L),P<0.05.Conclusions The expression of FHIT gene can increase the sensitivity of pancreatic adenocarcinoma to TNF.FHIT might be involved in the apoptotic path of TNF.
