反应停诱导骨髓瘤细胞株(KM3)凋亡的初步研究

Induction of Apoptosis by Thalidomide in KM3 Myeloma Cell Line

目的:观察反应停对KM3骨髓瘤细胞株的增殖抑制和诱导凋亡作用。方法:不同浓度的反应停作用于KM3细胞,光镜下计数细胞和胎盼蓝拒染法观察细胞增殖和细胞活力,细胞形态、细胞凋亡的荧光染色后的荧光显微镜检查、Annexin V-FITC和PI双染后流氏细胞仪分析判定凋亡和早期的凋亡细胞。结果:反应停在一定的剂量和一定的作用时间下可以抑制KM3细胞增殖,诱导KM3细胞凋亡,这一作用呈剂量和时间依赖性。结论:反应停诱导KM3细胞凋亡,抑制KM3细胞增殖可能是其直接抗骨髓瘤作用之一。

:To observe the effects of thalidomide on induction of cell apoptosis and inhibition of cell growth in KM3 cell line. Methods: Different concentrations of thalidomide were used to treat KM3 cells. Cell count and trypan blue exclusion by light microscope examination were used to observe the cell growth. Cell apoptosis and earlier apoptotic cells were investigated by cell marphology,apoptosis fluorescent assay and fluorescent activated cell sorter anaysis using Annexin V-FITC and Pistaining. Results:Thalidomide could inhibit KM3 cell growth and induce KM3 cell apoptosis. Within a certain range of treating time and dose, the effects depended upon time and dose. Conclusion: Thalidomide can induce KM3 cell apoptosis and inhibit KM3 cell growth.
