摘要
目的:观察健脾理气中药对肿瘤肝转移的抑制作用,探讨该方抗转移机制。方法:采用W256癌肉瘤细胞株脾下极接种造模。造模当天中药组动物每鼠灌喂蒸馏水溶解的健脾理气冲剂0.3ml(健脾理气中药浓度相当于生药剂量2g/ml),每日一次;对照组灌喂等量生理盐水,连续给药21d后处死动物,剥离脾脏瘤结节称重,计算抑瘤率;观察肝脏转移结节数,分离瘤组织,以原位杂交法检测瘤组织中MMP-1mRNA的表达。结果:健脾理气中药明显抑制脾脏移植瘤生长,中药组平均瘤重1.44±0.33g,抑瘤率达40.25%(p<0.001)。中药组肝转移灶数平均7.3±2.26个,转移抑制率为41.6%;肝转移灶瘤组织原位杂交检测结果显示:中药组MMP-1阳性表达率30%且无一样本呈强阳性,与对照组(70%)比较,差异显著(p<0.05)。结论:健脾理气中药能有效地抑制瘤细胞在肝脏的生长,防止肝转移的形成,该方抑制肝转移的作用与抑制瘤细胞MMP-1表达有关。
: To ascertain JH's effects on liver metastasis and discover its mechanisms. Methods: To replicate liver metastasis model, W256 carcinosarcoma cells were injected into lower part of Wistar rats, 2×106 cells for each rat. At the day of modeling, all rats were randomly divided into JH group and control group. Rats in the JH group were administrated orally 0.3ml JH once a day for 21 days; rats in control group were given equal volume of normal saline in same way. In the second day of experimental therapy, animals were killed, tumor nodules in spleens were incised, and tumor inhibitory rate were calculated; then, the number of metastatic clones on livers were counted. Lastly, MMP-1 mRNA in tumor tissues of metastases were detected by hybridization in situ.Results: JH can markedly inhibit transplanted tumor growth in spleen, the mean tumor weight in the JH group was 1.44±0.33 gram, with the tumor inhibitory rate of 40.25%(p<0.001 ). As there were only 7.3±2.26 metastatic clones in the JH group, its metastasis inhibitory rate reached as high as 41.6%. The results from hybridization in situ of metastasis tumor tissues showed that the MMP-1 expression rates in the JH group was 30% , without any intensive positive slices, which were significantly lower than that of the control group(p<0.05). Conclusion: JH can inhibit not only primary but secondary neoplasms, and down-regulation of MMP-1 expression may be its mechanisms.
