摘要
目的:检测基质金属蛋白酶2(MMP-2)和黏着斑激酶(FAK)在甲状腺乳头状癌中的表达,并探讨其相关性。方法:采用免疫组织化学EnVision方法,观察70例甲状腺乳头状癌及其癌旁正常组织中MMP-2和FAK的表达情况。结果:70例甲状腺癌组织中MMP-2和FAK的阳性表达率分别为84.3%(59/70)和80.0%(56/70);70例相应癌旁正常滤泡上皮中,阳性表达率分别为35.7%(25/70)和14.3%(10/70)。两者阳性表达率和阳性强度均以甲状腺乳头状癌中为高(P<0.05及P<0.01)。MMP-2分别在肿瘤直径大于或等于1cm组和伴淋巴结转移组中呈高表达,而FAK分别在40岁以上组和伴淋巴结转移组中呈高表达。MMP-2阳性表达强度与FAK的阳性表达强度之间呈显著正相关(P<0.01)。结论:甲状腺乳头状癌中MMP-2与FAK的表达密切相关,可作为甲状腺乳头状癌生物学行为预测的辅助指标。
: To detect the expression of matrix metalloproteinase-2(MMP-2) and focal adhesion kinase(FAK) in human thyroid papillary carcinoma, and to study their correlation. Methods: EnVision immunohistochemical technique was used to detect the expressions of MMP - 2 and FAK in 70 thyroid papillary carcinomas and adjacent follicular epithelia. Results: Positive expression rates for MMP-2 and FAK in thyroid papillary carcinoma were 84.3%(59/70) and 80.0%(56/ 70), and in the adjacent follicular epithelia to cancerous tissue were 35.7% (25/70) and 14.3% (10/70), respectively. Positive expression rates and levels of MMP - 2 and FAK in thyroid papillary carcinoma were significantly higher than those in the adjacent follicular epithelia(P < 0. 05, P<0.01, respectively). The expression of MMP - 2 was significantly higher in diameter of over 1cm and in cases with lymphnodes metastasis. The expression of FAK was higher in group of over 40 years and in cases with lymphnodes metastasis. There was a significant correlation between MMP - 2 and FAK expression(P<0. 01). Conclusion: The expression of MMP - 2 is related to the expression of FAK, which can be regarded as one of the parameters for evaluating the biological behavior of thyroid papillary carcinoma.
