摘要
目的 :建立稳定表达糖皮质激素受体β的细胞系。 方法 :构建糖皮质激素受体β的真核表达载体 ,用L ipotransfectamine法转染人骨肉瘤细胞系 HOS- 86 0 3,经 G4 18筛选后得到稳定转染细胞株 ;用极限稀释法获取亚克隆细胞株 ;用定量 RT- PCR方法鉴定糖皮质激素受体β m RNA的表达。结果与结论 :建立了稳定表达糖皮质激素受体β的细胞株 。
Objective:To establish a cell line expressing glucocorticoid receptorβ stably. Methods:The eukaryotic expression vector harboring glucocorticoid receptorβ c DNA was constructed and transfected into the human osteosarcom a cell line(HOS- 86 0 3) by the method of lipotransfectam ine. The stable transfectants were screened by G4 18. Cell subclones were isolated by utm ost dilution. The expression of m RNA of glucocorticoid receptorβ was determ ined by quantitative RT- PCR. Results and conclusion:A cell line stably expressing glucocorticoid receptorβ is established,and it can be used for the further biological study of the glucocorticoid receptorβ. [
