总合草苔虫中抗癌活性成分的提取和含量测定 被引量:4

摘要 目的 :建立总合草苔虫原料样品的预处理方法 ,用 RP- HPL C法测定其中抗癌活性成分草苔虫内酯含量。 方法 :采用 3种不同方法提取原料中草苔虫内酯 ,初提液经过硅胶柱和反相 ODS柱纯化 ,然后用 RP- HPL C法测定。结果 :二氯甲烷 -甲醇 ( 1∶ 1)超声提取液中草苔虫内酯含量较高 ;HPL C可以同时测定原料中的草苔虫内酯 4、5、10和 18,峰形和分离度良好。结论 :该方法可以方便地测定原料中的活性成分含量 ,为原料的质量控制奠定了基础。 Objective: To establish a method for detecting bryostatins in Bugula neritina L. by sample pre treatment and high performance liquid chromatography (HPLC). Methods: The efficiency of Soxhlet extraction and supersonic extraction was compared. Extracts from raw materials were successively purified through a gel silica and an ODS column, and then were assayed by RP HPLC. Results: Richer amount of bryostatins was obtained by supersonic extraction and lots of other constituents were eliminated by sample pre treatment. A good resolution of bryostatins 4,5,10 and 18 were obtained when assayed by HPLC. Conclusion: This method is suitable for assaying bryostartins in raw Bugula neritina L.,providing basis for raw material controlling.
