重组人内皮细胞抑制素腺病毒载体体外活性作用的研究

摘要 目的 :研究表达人内皮细胞抑制素 ( endostatin)的腺病毒载体的体外活性作用。 方法 :用重组人内皮细胞抑制素腺病毒载体体外转染人结肠癌 SW62 0细胞 ,观察肿瘤细胞生长情况 ,并观察其上清对大鼠肺毛细血管内皮细胞、新生牛主动脉内皮细胞及人脐静脉内皮细胞生长的影响。 结果 :MTT法检查证实重组腺病毒对结肠癌细胞生长无抑制 ,锥虫蓝染色证实转染的人结肠癌 SW62 0细胞上清对各种血管内皮细胞的生长均有较强的抑制作用。 结论 :重组人内皮细胞抑制素腺病毒载体体外对各种血管内皮细胞生长均有较强的抑制作用 。 Objective:To study the activity of adenovirus vector expressing human endostatin in vitro .Methods:The colon cancer line SW620 was infected with the recombinant adenovirus.The growth of the cells was detected,and the activity of the supernatant of infected SW620 cells on the growth of RVEC,BAEC and HUVEC was explored. Results: Western blot indicated endostatin protein was expressed in the supernatant.MTT indicated no inhibition on the growth of cancer cells infected with adenovirus vector expressing human endostatin. Trypan blue staining indicated the supernatant had strong inhibitory effect on the growth of 3 kinds of vascular endothelial cells. Conclusion: Adenovirus vector expressing human endostatin has strong inhibitory effects on the growth of vascular endothelial cells from different kinds of species,and can be used in animal study.
