大鼠睡眠剥夺后行为及下丘脑诱生型一氧化氮合酶mRNA表达的变化 被引量:14

摘要 目的 :观察快动眼睡眠 ( rapid eye movement sleep,REMS)剥夺大鼠行为的改变及其下丘脑诱生型一氧化氮合酶( i NOS) m RNA表达的时相变化。方法 :小站台水环境法剥夺大鼠睡眠 2 4、4 8、72 h后 ,用旷场实验 ( open field test,OFT)观察大鼠主动行为的变化 ,同时用半定量逆转录聚合酶链反应 ( RT- PCR)检测其下丘脑 i NOS m RNA的表达。结果 :睡眠剥夺2 4、4 8h组大鼠的旷场实验得分较非剥夺组高 ( P<0 .0 1) ,剥夺 72 h后降低 ;睡眠剥夺 2 4 h及 4 8h组 i NOS m RNA的表达量无显著变化 ,但剥夺 72 h组 i NOS m RNA的表达量大幅度增加 ( P<0 .0 1)。结论 :内源性一氧化氮 ( NO)增加可能是睡眠剥夺后机体精神行为变化的重要原因 ,高水平的 Objective: To study the effects of rapid eye movement sleep (REMS) deprivation on active behavior and the expression of mRNA coding for inducible nitric oxide synthase (iNOS) of hypothalamus in rats. Methods: The flower pot technique was adopted to deprive the REMS of Sprague Dawley rats for 24 h, 48 h and 72 h respectively. The active behavior of rats were examined by the open field test (OFT). The expression of mRNA coding for iNOS of hypothalamus in rats was assayed by reverse transcription polymerase chain reaction (RT PCR). Results: The OFT scores were significantly increased in rats subjected to 24 h and 48 h REMS deprivation ( P <0.01), but the score in 72 h deprivation group was decreased and not significantly different from that in the control group ( P >0.05). There was no difference in the expression of iNOS mRNA among 24 h, 48 h REMS deprivation and the control groups, but the expression was significantly increased in 72 h REMS deprivation group ( P <0.01). Conclusion: Excessive nitric oxide might be a major factor for the changes of neuronal behavior caused by sleep loss directly or indirectly by other sleep factors.
