人参皂甙对白血病耐药细胞化疗药物敏感性的研究 被引量:4

摘要 目的 :通过观察人参皂甙 ( GS)协同化疗药物对 K5 62 /VCR、K5 62 /Adr白血病耐药细胞株的抑制作用 ,探讨 GS是否能提高肿瘤耐药细胞对化疗药物的敏感性。方法 :选用 K5 62 /VCR、K5 62 /Adr两耐药细胞株作为靶细胞 ,液体培养和半固体集落培养法观察不同浓度的 GS对耐药细胞增殖的影响 ,及 GS协同化疗药物对 K5 62 /VCR、K5 62 /Adr耐药细胞集落生成的抑制作用。结果 :1单用 GS作液体培养 MTT分析法和集落培养法均显示低、中等浓度的 GS5~ 5 0 μg/ml对细胞增殖无明显影响 ,但当浓度升高至 75 μg/ml时 ,则抑制细胞增殖和集落形成 ( P<0 .0 1 )。2当 GS与化疗药物联合应用时 ,即使化疗药物浓度 ( 1 μg/ml)不变 ,K5 62 /VCR、K5 62 /Adr耐药细胞对化疗药物的敏感性与 GS呈剂量依赖关系。即 GS浓度达到 5 0μg/ml时 ,化疗药物对细胞的抑制作用明显 ( P<0 .0 1 ) ,且随着 GS浓度的升高 ,抑制作用逐渐增强。结论 :GS通过提高耐药细胞对化疗药物的敏感性而与化疗药物起协同作用 ,同时在一定浓度下 。 Objective: To explore the effects of Ginsenosides (GS) on leukemia cell lines resisted to cytotoxic drugs. Methods:The leukemia cell lines of K562/VCR K562/Adr resisted to cytotoxic drugs were used as targets cells, the effects of GS alone and the synergy with cytotoxic drugs were observed by MTT method and colony forming unite assay in vitro. Results: ① The growth of K562/VCR and K562/Adr cells were inhibited by higher concentration of GS (75~100μg/ml), but not suppressed by low or medium concentration of GS (5~50μg/ml) both in liquid and semi solid culture system.②In the combination of GS with cytotoxic drugs, K562/VCR and K562/Adr cells became more sensitive to low concentration of cytotoxic drugs (1μg/ml of VCR or Adr), and inhibited obviously by GS in the manner of dose dependence. Conclusion:GS could enhance susceptibility of leukemia cells to cytotoxic drugs from resistance to response, and meanwhile inhibit these cells in the presence of its higher concentrations.
