摘要
目的 :探讨BALB/C和C5 7BL/6小鼠创伤反应差异性的分子遗传学机制。方法 :利用基因芯片技术对BALB/C和C5 7BL/6小鼠在创伤后 1h和 6h肝组织基因表达谱进行比较研究 ,对在两种小鼠创伤反应早期差异表达的基因进行聚类分析。结果 :对 132 7条差异表达的基因进行聚类分析后发现有两大族基因的表达在BALB/C小鼠和C5 7BL/6小鼠创伤后早期存在明显差异。BALB/C小鼠糖类代谢、脂类代谢和激素代谢活跃 ,细胞信号转导以G蛋白信号通路和Ca2 + 信号通路激活为主 ,而且与组织损伤相关的基因也出现高表达。而C5 7BL/6小鼠急性时相反应和免疫反应被激活 ,细胞凋亡途径被抑制 ,细胞信号转导以酪氨酸蛋白激酶系统激活为主要特征。结论 :BALB/C和C5 7BL/6小鼠的创伤反应差异性可能与它们在创伤后基因组的表达方式不同相关。
Objective:To investigate the molecular genetic mechanisms of posttrauma-response heterogeneity between BALB/C and C57BL/6 mice.Methods:The hepatic gene expression patterns were analyzed by gene chips (cDNA microarray) at 1 and 6 hours after blast injury,and the differential expression genes were clustered.Results:Cluster analysis identified two distinct groups of genes involved in some physiological functions.In BALB/C mice there were many enzymes and associated factor genes involved in metabolism of sugar,lipid and hormones up regulated after injury,and the G protein and Ca 2+ signal pathways were activated,the over expression of tissue and cell injury associated factor genes were observed in the meantime.On the other hand,the characterizations of C57BL/6 mice showed the acute phase reaction and immunoreaction were activated and the apoptosis were inhibited in the early phase after injury.A rapid elevation of gene transcription in the tyrosine kinase receptors signal pathways were observed also.Conclusions:The differential genome expression profile between BALB/C and C57BL/6 mice maybe contribute to their posttrauma-response heterogeneity after blast injury.
