氧化苦参碱与国产α1b-和α2b-干扰素对大鼠肝星状细胞增殖和胶原合成的抑制作用( 英文) 被引量:8

摘要 目的 研究氧化苦参碱与国产α 1b -和α 2b -干扰素对大鼠肝星状细胞增殖和胶原合成的影响 ,为应用氧化苦参碱防治肝纤维化提供理论依据。方法 分离、培养大鼠肝星状细胞 ,采用MTT法和3 H -脯氨酸掺入法分别测定细胞增殖和胶原合成。结果 氧化苦参碱在 5 μg~ 5 0 0 μg/mL ,两种α -干扰素在 5 0 0U -10 0 0 0U/mL浓度范围内能呈剂量依赖性抑制星状细胞的增殖和胶原合成 ,提高药物浓度则形成作用平台。结论 氧化苦参碱与α -干扰素在体外均具有抑制大鼠肝星状细胞增殖和胶原合成的作用 。 Aim To investigate the effect of oxymatrine,a chinese herbal medicne,and interferon α 1b and α 2b on proliferation and collagen synthesis of rat stellate cells in vitro to provide theary foundation for using oxymatrine to treat live fibrosis.Method Isolate and culture rat hepatic stellate cells,and the cell proliferation and collagen synthesis were tested by MTT and 3H proline incorporation,respectively.Results The inhibition of proliferation and collagen synthesis by oxymatrine at dose of 5 μg 500 μg/mL,and by interferon α 1b and interferon α 2b at 500 U-10 000 U/mL were observed to be in a concentration dependent manner.Conclusions The Chinese herbal medicine,oxymatrine,has the similar inhibiting potential as interferon α on rat stellate cells in vitro.
