大鼠隐睾固定后生殖细胞发育凋亡与iNOS基因表达

摘要 目的 :研究大鼠隐睾固定后生殖细胞的发育和凋亡 ;探讨诱生型一氧化氮合酶 (iNOS)基因表达与生殖细胞发育和凋亡的关系。方法 :SD雄性大鼠 ,日龄 2 2d时复制双侧隐睾模型 ,术后 14d复制隐睾固定模型。用生物素 dUTP 酶标亲和素测定法检测生殖细胞凋亡 ;用免疫组化SP法检测iNOS基因表达。结果 :与对照组相比 ,隐睾组发生凋亡的生殖细胞数显著增加 (P <0 .0 1) ,隐睾固定组发生凋亡的生殖细胞数无显著变化 (P >0 .0 5 )。发生凋亡的生殖细胞胞浆中iNOS基因表达明显增强。结论 :手术建立的隐睾生殖细胞凋亡增加。隐睾固定后生殖细胞凋亡减少。 Objectives: To study the effect of orchidopexy on germ cell development and apoptosis, and explore the relationship between iNOS expression and germ cell apoptosis. Methods: Immature rats were subjected to bilateral cryptorchidism, and 14 days later and orchidopexy was performed on a subset of these rats.Germ cell apoptosis was detected by terminal deoxynucleotidyl transferase mediated d UTP nick end labeling (TUNEL). Expression of iNOS were detected by immunohischemical SP method. Results: The number of apoptotic germ cell in cryptorchid testes increased significantly compared with the control (P<0.01). By contrast, the number of apoptotic germ cell in orchidopexied testes was not predominatly different from that of the control (P>0.05). Intensive expression of iNOS was detected in the cytoplasm of degenerated germ cell. Conclusions: Cryptochidism can induce germ cell apoptosis and orchidopexy can lower the level of germ cell apoptosis. Expression of iNOS is closely assoiated with germ cell development and apoptosis.
