摘要
目的 :构建抗人乳腺癌单克隆抗体人鼠嵌合抗体 ,降低鼠源性抗体应用于人体的免疫排斥反应。方法 :采用 RT- PCR技术 ,从分泌抗人乳腺癌单克隆抗体的杂交瘤细胞株 CDI 315 B分离克隆抗体轻重链可变区基因 ,对其进行序列分析。将连接组建人鼠嵌合轻链和人鼠嵌合重链 ,然后分别将它们克隆到真核细胞表达载体 pc DNA3。结果 :构建的人鼠嵌合抗体基因克隆在真核细胞表达载体上。结论
Objective: To construct an anti breast cancer human mouse chimeric antibody and reduce its human anti mouse antibody response in vivo application Methods: The entire VH and VL genes of anti breast cancer McAb CDI 315B were amplified by RT PCR Mouse VH and VL genes were joined to human Cr1 gene and human CL gene and the recombination genes were subsequently subcloned into expression vector pcDNA3 Results: Chimeric antibody was constructed in an eukaryotic expression vector pcDNA3 Conclusion: This chimeric antibody will be further expressed in mammalian cells and provide a new approach to utilize human mouse chimeric antibody in clinical treatment of patients with breast cancer
