摘要
目的 :研究大鼠局灶性脑缺血不同缺血时间皮质半暗带和中心区淀粉样前体蛋白 (APP)在转录水平表达规律。方法 :用线栓法建立大鼠局灶性脑缺血模型 ,剥取缺血半暗带及中心区皮质组织 ,采用半定量逆转录 聚合酶链式反应 (RT PCR) ,测定APPmRNA水平的变化。结果 :半暗带APPmRNA在缺血后 48h升高 ,缺血 72h达到高峰 ,缺血 1周后仍高于正常。缺血中心区APPmRNA在缺血后 72h和 96h高于正常水平。结论 :APPmRNA在缺血半暗带的表达上调 ,有可能加重缺血损伤。
Aim:To investigate the changes of amyloid precursor protein(APP) transcriptional level at different ischemic times in rats' ischemic penumbra and core of cortices.Methods:Focal ischemic models of middle cerebral artery occlusion (MCAO) were achieved by inserting nylon surgical thread through the internal carotid artery (ICA) into the anterior cerebral artery(ACA). Brain samples were harvested from ischemic penumbra and core of cortices. The expression of APP mRNA was assessed by RT PCR. Results:APP mRNA in the penumbra regions enhanced in 48 h after ischemia and reached a peak at 3 day. In 1 week following ischemia, it was still higher than normal level. As for ischemic core regions,APP mRNA increased at 72 h and 96 h following ischemia. Conclusion:APP mRNA expression was up regulated in the penumbra regions after focal cerebral ischemia. It might aggravate ischemic injury.
