奥曲肽对肝部分切除术后大鼠肝再生影响的实验研究

The influences of Octreotide on rat liver regeneration undergone partial hepatectomy

目的 了解生长抑素 8肽类似物奥曲肽对肝部分切除术后大鼠肝脏再生和肝功能的影响 ,探讨奥曲肽手术后应用的安全性。方法 将 80只♂成年Wistar大鼠随机分为 5组 ,每组 16只。A、B组为单纯左肝切除组 ,其中B组腹腔注射奥曲肽 (5 0 μg/kg、Bid) ;C、D、E组均为左肝切除加右肝肿瘤种植组 ,D、E组分别在肿瘤种植后 12、72h开始同样方法剂量应用奥曲肽 ;A组为空白对照、C组作为荷瘤对照。结果 术后 1周A、B组间及C、D、E组间肝脏重量、增殖细胞核抗原 (PCNA)表达和DNA增殖以及术后肝功能等差异没有显著性 (P >0 0 5 )。结论 生长抑素类似物奥曲肽对部分切除后大鼠肝脏再生没有影响 ,并且与是否荷瘤无关。

Objective To assess the influences and security of Octreotide on rat liver regeneration and liver function undergone partial hepatectomy. Methods 80 male adult Wistar rats were divided into 5 groups, 16s in each group. Group A and B took only left lobe partial hepatectomy while Ocreotide (50 μg/kg)were administered intraperitoneally to group B twice a day. Walker-256 tumor was implanted in right lobe of liver of group C, D, E when their partial hepatectmoy of left lobe were operated. The first administered time for Octreotide on group D or E was 12 h and 72 h after resection respectively. Group A and C acted as blank and carrying control received a similar volume of saline. Results Each rat in weight of remnant liver, expression of PCNA, regenerative hepatic cell DNA content and liver function one week after resection were analyzed. There is no significant difference between A versus B or C, D versus E. Conclusion Octreotide has no significant suppression on hepatic cell regeneration without any relation with carrying tumor or not.
