香菇多糖对荷瘤小鼠肝组织的干扰素和超氧化物歧化酶 m RNA 表达 被引量:7

Effect of Lentinal on the Expression of IFN mRNA and SOD mRNA in S180-bearing C57 BL/6 Mice

摘要 目的 :研究香菇多糖 (L NT)对荷瘤小鼠肝组织中干扰素 (IFN)和超氧化物歧化酶 (SOD)基因 (m RNA )转录的影响。方法 :生长正常的 C57BL /6小鼠 ,左腋皮下接种 S1 80 细胞 ,接种 4 8h后 ,分荷瘤组、荷瘤加低 L NT组(浓度为 5 mg/kg)、荷瘤加高 L NT组 (浓度为 30 m g/kg)及对照组 ,每组 4只 ,连续给药 6 d后 ,进行断颈处死 ,剖取瘤体称重记录和提取鼠肝组织总 m RNA斑点杂交。结果 :L NT对 S1 80 肉瘤的生长有较明显的抑制作用 ,而 5 m g/kg和 30 m g/kg两种剂量的 L NT均能提高 IFN和 SOD m RNA的表达量 ,且前者作用稍强于后者 (P <0 .0 5 )。结论 :L NT能提高荷瘤小鼠肝组织 IFN和 SOD的 m Objective: To study the effect of lentinal (LNT) on the expression of IFN mRNA and SOD mRNA in S180 bearing C57 BL/6 mice.Methods: S180 cells were implanted into the axilla of normally growing C57 BL/6 mice by subcutaneous injection. After 48 h, the mice were divided into three groups (n=4 each): HT (Holland Tumor) group, HT with lower LNT (5 mg/kg) group and HT with higher LNT (30 mg/kg) group. After the drug was given continuously for 6 days, the mice were killed by decapitation. The tumors were removed and weighted. Total mRNA in the liver tissue of the mice was extracted for blot hybridization.Results:LNT could obviously inhibit the growth of S180 sarcoma. Both dosages of 5 mg/kg and 30 mg/kg of LNT could increase the expression of IFN mRNA and SOD mRNA with the former effect being slightly stronger than the latter (P<0.05).Conclusion:LNT can enhance the expression of IFN mRNA and SOD mRNA in S180-bearing C57 BL/6 mice.
