梯度离心法在肿瘤药物敏感性试验中的应用研究

Application of gradient centrifugation for the trial of the anti-cancer drug's sensitivity

目的 研究梯度离心法在肿瘤细胞药物敏感性试验中的应用。方法 采用淋巴细胞分离液梯度离心法分离纯化癌细胞 ,MTT比色法测定 4 5例体外原代癌细胞对 8种常用化疗药物的敏感性。结果 采用梯度离心法收集纯化癌细胞 ,癌细胞获得百分率为 94 .6 5± 2 8.4 7,药敏阳性率为 71.11% ,与传统方法相比 ,差异有非常显著性 (P<0 .0 1)。结论 梯度离心法 MTT肿瘤体外药敏试验是一种良好的实验方法 。

Objective To study the application of the gradient centrifugation for the trial of anti cancer drug's sensitivity.Methods Cancer cells were gradient centrifugalized and purfied and then the sensitivity in 45 cases of originative cancer cells to 8 kinds of common chemotherapeutic drugs was detected in vitro by MTT colormetric assay.Results The percentage of cancer cells collected by gradient centrifugalization was 94.65±28.47,positive rate of drugs sensitivity was 71.11%,which is significantly higher than those of traditional methods( P <0.01).Conclusion Gradient centrifugalization with MTT colorimetric assay is a better way in experimental drugs sensitivity,which is worth applying in clinical routine works.
