氧化砷诱导食管癌细胞凋亡线粒体超微结构和膜电位的改变 被引量:4

Ultrastructural and Transmembrane Potential of Mitochondrial inApoptotic Esophageal Carcinoma Cell Induced by Arsenic Trioxide

摘要 目的 :研究氧化砷 (As2 O3)诱导食管癌细胞凋亡的过程中线粒体超微结构和功能的变化。方法 :培养的食管癌细胞系SHEE85加入 3 μmol LAs2 O3,作用 2 ,4 ,6,12 ,2 4h ,在光镜和电镜下观察细胞形态。以罗丹明 (Rhodamine 12 3 )荧光探针标记线粒体 ,细胞荧光光度计检查线粒体荧光强度 ,计算线粒体跨膜电位 (MTP ,△Ψm)的改变。结果 :线粒体荧光强度的改变反映MTP的状态。荧光强度早期增加 ,中期下降 ,后期最低 ,2 4h后细胞核呈典型凋亡改变伴有线粒体肿胀。结论 :在As2 O3诱导下 ,SHEE85细胞线粒体有明显的形态和功能改变伴随线粒体跨膜电位的降低 ,线粒体改变是As2 Objective:To study the ultrastructural and functional change of mitochondria in the process of apoptosis of esophageal carcinoma cell induced by arsenic trioxide (As 2O 3). Methods: The cultural esophageal carcinoma cell line SHEE85 was treated with As 2O 3(3μmol/L). After 2, 4, 6, 12, 24 hours of drug adding, the SHEE85 cells were collected for light and electron microscopic examination. The mitochondria were labeled by Rhodamine 123 fluorescence probe and the fluorescent intensity of the mitochondria was measured by fluorescent microphotometry. Further the mitochondrial transmembrane potential (MTP,△Ψm) changes were also calculated. Results: The functional changes of the mitochondria calculated by fluorescent intensity, which reflected the MTP status of mitochondria. The fluorescent intensity increased in early stage, declined in mid stage and decreased to the lowest in the late stage. After 24 hours of adding As 2O 3, the cell nucleus showed the typical apoptotic changes accompanying swelled mitochondria. Conclusion: Under the inducement of As 2O 3,SHEE85 cells display the apparent morphological and functional changes of mitochondria with decrease of mitochondrial transmenbrane potential. Therefore, the change of mitochondria is an important intermediate link in the course of apoptosis of esophageal carcinoma cells induced by As 2O 3.
