合成肽检测抗α_1-肾上腺素受体自身抗体方法及其临床意义 被引量:2

摘要 目的 建立抗α1 肾上腺素受体 (anti α1 AdR)自身抗体的ELISA ,评价其与高血压的关系。方法 以合成的α1 肾上腺素受体细胞外第二环功能表位肽段 (α1 192~ 2 18位氨基酸 )作抗原 ,建立间接酶联免疫吸附试验 (ELISA) ,检测 98例难治性高血压、96例非难治性高血压患者和 4 0例正常人血清中anti α1 AdR。结果 阳性参考血清批内变异系数 (CV)和批间CV分别为 0 0 72、0 10 1。用抗原吸收后 ,吸光度 (A)值降低 1 3倍。 98例难治性高血压患者中 ,anti α1 AdR的阳性率为 36 7%(36 / 98) ,明显高于非难治性高血压组 13 5 4 % (13/ 96 )和正常血压对照组 5 0 % (2 / 4 0 )。结论 检测抗α1 肾上腺素受体自身抗体的方法特异性和敏感性较高 ,操作简便 。 Objective Establish a method to screen the autoantibodies against α 1 adrenergic receptor by ELISA and to assess its value for hypertension.Methods The epitope on peptide of the 2 nd extracellular loops(amino acid sequence from 192 to 218)of α 1 adrenergic receptor was synthesized and used as antigen to screen the autoantibodies by ELISA.The antoantibodies in 98 patients with refractory hypertension and normal persons were assayed.Results The intra and inter assay CV s of were 0.072 and 0.101,respectively,There was a 2.5-fold decrease in A value after absorption of antigen.The autoantibodies against the α 1 adrenergic receptor were found in 36 of the 98(36.7%)patients.In control group,the positive rate was 5 0%.There was a significant difference between the patient′s group and control group.Conclusion This is an simple,specific,sensitive and stable for screening the anti α 1 adR. The method autoantibodies in hypertensive patients.
