金黄色葡萄球菌随机扩增多态性DNA分型及其应用 被引量:8

摘要 目的 利用随机扩增多态性DNA(randomamplifiedpolymorphicDNA ,RAPD)分析技术 ,对金黄色葡萄球菌进行基因分型 ,并观察临床耐药型与其基因型之间的关系。方法 利用自己合成的随机引物 ,建立RAPD检测分析方法 ,并利用这一方法对 5株金黄色葡萄球菌进行基因分型的研究。结果  5株金黄色葡萄球菌中 ,2株耐甲氧西林金黄色葡萄球菌 (MRSA)为同一基因型 ,另外 3株为两种基因型 ,均是敏感菌株。结论 从RAPD分析的初步结果可以看出 ,金黄色葡萄球菌的临床表现型与基因型存在一定的关系 。 Objective Using genotyping method to analysis the relationship between the phenotype and genotype of drug resistance to antibiotics.Method Random amplified polymorphic DNA(RAPD)technique was used to analysis both sensitive and resistant Staphylococcus aureus strains which were previously isolated from the samples of infected patients.Result Five strains can be identified into three genotypes.And among these three,there is a genotype of methiclillin-resistant Staphylococcus aureus (MRSA).Conclusion RAPD is a very sensitive genotyping method.Primary result shows that MRSA is related to a certain genotype of Staphylococcus aureus .It may be used to do the quick identification of MRSA on the clinical samples directly without any further bacterial culture procedure.
