摘要
目的 探讨两种血液肿瘤细胞K5 6 2和HL 6 0培养上清液对脐带血CIK细胞的影响作用。方法 在细胞因子诱导的脐带血杀伤细胞 (CIKs)形成的过程中 ,加入肿瘤细胞培养上清 ,观察CIK细胞的生长情况 ,并同正常培养条件下的CIK细胞比较增殖倍率、免疫表型及杀伤活性。结果 脐带血CIK细胞是以CD3+ T细胞为主的异质性细胞群体。随培养时间延长 ,CD3+ CD5 6 + 细胞比例明显升高。K5 6 2和HL 6 0培养上清液能明显抑制CIK的增殖率和杀伤活性 ,免疫表型亦有显著的变化 (P <0 0 5 )。结论 K5 6 2和HL 6 0培养上清液能抑制脐带血CIK细胞的活性 。
Objective To investigate the effects of cultural supernanant of tumor cells on cytokine induced killer cells(CIKs) in vitro in cord blood.Methods CIKs were derived from cord blood monocytes.The cultural supernanants of K562 and HL 60 cells were added and the growth of CIKs was observed.The CIKs in routine culture were used as contrals.The cytotoxicity against K562 and HL 60 target cells was measured by MTT method.Immune phenotypes of CIKs were analyzed by FACS.Result Cord blood CIKs were heterogenic cell population consisting of mainly CD3 +T cells.The ration of CD3 +CD56 + cells increased as culture continued.CIK added with the supernanant of tumor cells exhibited some defects over CIK alone in cytotoxicity,immunotypes,proliferation( P <0 05).Conclusion The activity of cord blood of CIKs were inhibited by supernanant of tumor cells.
