bFGF反义寡核苷酸抑制大鼠实验性晶状体损伤后晶状体上皮细胞的增殖和迁移 被引量:4

摘要 目的 :探讨bFGF的反义寡核苷酸对大鼠实验性、外伤性白内障中晶状体上皮细胞增殖和迁移的作用。方法用注射器经角膜将大鼠双眼晶状体的前囊膜刺破并刺入皮质中 ,造成外伤性白内障。然后将 12只动物分为 3组 ,前房分别注射生理盐水、0 .1%反义寡核苷酸和 0 .1%正义寡核苷酸 ,在第 3、7、10和 14天取出眼球 ,用 10 %甲醛溶液固定 ,行病理切片、HE染色 ,在显微镜下观察晶状体上皮细胞的增殖、迁移及赤道部细胞和细胞空泡。结果 :晶状体的组织学显示 ,反义寡核苷酸在第 3、7和 10天在抑制晶状体上皮细胞增殖及迁移方面的作用强于生理盐水及正义寡核苷酸。在赤道部细胞活跃方面 :生理盐水对照组存在 3天 ,正义寡核苷酸存在 2天 ,而反义寡核苷酸没有。在细胞空泡方面 :生理盐水对照组有 2天存在 ,而正义寡核苷酸和反义寡核苷酸组则不存在。统计学分析显示 ,反义寡核苷酸组在抑制晶状体上皮细胞增殖方面的作用显著强于生理盐水组 (P =0 .0 10 4 )和正义寡核苷酸组 (P =0 .0 4 99) ,在抑制晶状体上皮细胞向后囊迁移方面显著强于生理盐水组 (P =0 .0 0 11) ,而与正义寡核苷酸组间差异无显著性意义 (P =0 .0 830 )。在赤道部细胞活跃 (P =0 .4 317)和细胞空泡方面 (P =0 .6 188) ,三组间差异无显著性意义。结论 :? Objective:To evaluate the effects of bFGF antisense oligonucleotides on the proliferation and migration of lens epithelial cells in experimental rat traumatic cataract.Methods:Syringes inserted through the cornea were used to puncture the anterior capsules of rat lenses and rat traumatic cataract was induced. Rats were divided into three groups. The anterior chambers were injected with natural saline,0.1% antisense oligonucleotides or 0.1% sense oligonucleotides. The eyeballs were retrieved on the third,seventh,tenth and fourteenth days. They were fixed with 10% formaldehyde and mounted on slides. They were dyed with HE,then proliferation and migration of lens epithelial cells,equator cells,and cell vacuoles were observed under microscope.Results:In histology,the antisense oligonucleotides had a greater effect on proliferation and migration than contrast and sense oligonucleotides on the third,seventh and tenth days. In equator cell activity,the contrast group could be observed for 3 days,the sense group could be observed for 2 days and the antisense group was not evident. For cell vacuoles,the contrast group could be observed for 2 days but the sense group and the antisense group were not evident. Statistically,the antisense oligonucleotides had significantly greater effects on proliferation than the natural saline(P=0.0104) or sense oligonucleotides(P=0.0499). It had significantly greater effects on migration than the natural saline(P=0.0011) but there was no significant difference with the sense oligonucleotide(P=0.0830). There was no significant difference on equator cell activity(P=0.4317) and cell vacuoles(P=0.6188) among the three groups.Conclusion:Basic fibroblast growth factor antisense oligonucleotides had inhibiting effects on proliferation and migration of lens epithelial cells in experimental rat traumatic cataract.
