流式细胞仪检测非小细胞肺癌中CD44的表达及凋亡细胞峰 被引量:2

摘要 目的 研究粘附分子CD4 4在细胞凋亡中的作用 ,探讨CD4 4在非小细胞肺癌发生、发展及转移中的作用。方法 取新鲜NSCLC标本 2 8例 ,应用流式细胞术测定CD4 4 ,同时测定凋亡细胞峰 ,并与癌旁肺组织进行比较。结果 肿瘤组织和癌旁肺组织CD4 4的表达分别为 :5 7.3± 3.74 %和 7.14± 1.0 1%。两者比较差异极其显著 (P <0 .0 1)。凋亡细胞峰的测定 ,肿瘤组织为 14 .4 6± 2 .18% ,癌旁肺组织为 0 .75± 0 .1% (P <0 .0 1)。肺癌组及正常肺组织CD4 4和凋亡细胞峰的表达均具有相关性 (分别为Rs=0 .2 7,P <0 .0 5和Rs =0 .5 5 ,P <0 .0 1)。结论 CD4 4能抑制T细胞对凋亡的免疫调节功能 ,使癌细胞逃避机体免疫机制 ,促进肿瘤的生长和转移。当CD4 4的表达和功能异常时 ,影响细胞内凋亡相关基因 ,使得正常的细胞凋亡障碍 。 Objective To study the expression of adhesion molecule CD44 in non-small cell lung carcinoma and to determine the role of CD44 in tumor genosis and progress. Methods With flow cytometry, expression of CD44 and apoptotic cell peak were detected in 28 fresh specimens from NSCLC and adjacent normal pulmonary tissues. Results In NSCLC and normal pulmonary specimens, expressions of CD44 were 57.3±3.74% and 7.14±1.01%,apoptotic cell peaks were 57.3±3.74% and 7.14±1.01% ( P <0.01). A good correlation was observed between the expression of CD44 and apoptotic cell peak in NSCLC and normal pulmonary specimens(Rs=0.27, P <0.05 and Rs=0.55, P <0.01). Conclusion CD44 may play an important role in restraining the apoptosis regulating function of T cell, and thus tumors can escape from body's immunization mechanism. The disturbance of expression of CD44 may infuence apoptosis, and may lead to hyperproliferation and malignant change of cells, this may be one of the causes of lung carcinoma.
