摘要 目的 研究 p2 7基因对人胃癌细胞SGC 790 1的抗增殖作用。方法 构建重组体腺病毒Ad p2 7kip1并转染SGC 790 1细胞 ,绘制细胞生长曲线 ,克隆形成实验 ,免疫组化检测PCNA的表达情况。 结果 成功构建Ad p2 7kip1 ,病毒滴度为 1 .2 4× 1 0 12 pfu/ml,在MOI≥ 5 0时 ,即可达到 1 0 0 %的转导效率。在SGC 790 1细胞中表达 ,能抑制其生长和集落形成 ,Ad p2 7kip1和对照组PCNA标记指数分别为 3 5 .2 %及78.6% (P <0 .0 1 )。结论 p2 7可有效抑制胃癌细胞的增殖活性 ,这为采用 p2 7kip1cDNA基因治疗胃癌奠定了基础。 Objective To study the effects of p27kip1gene on the proliferation in gastric cancer cell lines SGC\|7901. Methods A replication\|deficient adenovirus vector encoding p27kip1(ad\|p27kip1) was constructed and human gastriccancer cell lines SGC\|7901were transduced in vitro. Analyse the growth curve,colne formation experiment, and examine the PCNA by immunohistochemistry.Results Ad\|p27kip1 containing the p27kip1 sequence was successfully constructed and the virus titer was 1.24×10 12 pfu/ml.The transduction efficiency was 100% when MOI ≥50. The growth was inhibited and the colne was reduced in ad\|p27kip1\|transduced gastriccancer cell lines. The difference of PCNA expression in ad\|p27kip1 group and in control group was statistically significant ( P <0.01).Conclusion p27kip1 gene could inhibit the proliferation of gastric cancer cell lines SGC\|7901, This study provides the basis for future gastric cancer treatment based on p27kip1cDNA strategy.
