摘要
目的 建立一种简便可行的RP—HPLC法测定肾移植术后病人服用环孢素A(CsA)后全血中的药物浓度 ,为临床合理应用CsA提供一个良好的指标。方法 病人血样经乙醚单步提取后 ,以C8硅胶键合相为固定相 ,乙腈 -甲醇 -水 (40∶34∶2 6 )为流动相 ,6 0℃柱温下进行色谱分析 ,紫外检测波长为 2 14nm。结果 CsA全血浓度在 5 0~ 6 0 0ng·ml-1范围内呈良好的线性关系 ,r =0 .9998,方法的平均回收率为 96 .0 8% ,日内和日间精密度RSD均小于 6 % (n =5 ) ,最低检测限为 10ng·ml-1。结论 本法简便、快速、易于开展 ,用于肾移植术后患者CsA的血药浓度监测 。
Aim To set up a practical RP HPLC method to determine the concentration of cyclosporin A(CsA) in patients' whole blood after renal transplantation.Methods CsA was extracted from whole blood with diethyl ether and separated using a C 8 column and a mobile phase of acetonitrile methanol deionized water(40∶34∶26). The column temperature was maintained at 60℃.CsA was detected at 214nm.Results A good linear relationship was obtained between the amount of CsA from 50ng·ml -1 to 600ng·ml -1 ,r=0.999 8.The average recovery rate of CsA was 96.08%,and the RSD within day and between day were less than 6% (n=5).The detection limit of CsA was 10ng·ml -1 .Conclusion The established method was accurate,simple,sensitive and suitable for the therapeutic monitoring of CsA.
