摘要
目的:探讨β-榄香烯对大鼠胃粘膜癌变发生、发展过程中bcl-2基因mRNA表达及端粒酶活性的影响。方法:采用甲基硝基亚硝基肌(MNNG)、10%NaCl混合液加40%乙醇灌胃建立大鼠胃粘膜癌变模型,设立正常组、模型组及榄香烯组,榄香烯组在造模过程中采用β-榄香烯干预。30周后,分别采用原位杂交及端粒微孔板杂交法检测大鼠胃粘膜bcl-2基因mRNA表达及端粒酶活性。结果:榄香烯组癌变发生率低于模型组。在大鼠胃粘膜癌变的发展过程中,bcl-2基因mRNA表达与端粒酶活性呈递增趋势,且两者具高度相关性。结论:下凋bcl-2基因表达抑制端粒酶活性,是β-榄香烯重要抗癌机制之一。
Background/Aims: To study the effects of J3-elemene on bcl-2 mRNA gene expression and telomerase activity of gastic cancer in Wistar rats. Methods: We studied the expression of bcl-2 gene mRNA and telomerase activity using in situ hybridization in rats of three groups: control group, model group and elemene group. In the model group gastric cancer was induced by MNNG, 10% sodium chloride, and 40% ethanol. In the elemene group gastric cancer was induced by MNNG and elemene was given at the same time. Results: The occurrence rate of gastric cancer in the elemene group was lower than that of the model group. Along with the development of gastric cancer, the expression of bcl-2 gene mRNA and telomerase activity in gastric mucosa was highly correlated and had a trend to increase. Conclusions: β-elemene can reduce the expression of bcl-2 gene mRNA and inhibit telomerase activity in gastric cancer.
