影响人参发根中皂甙含量基因的定位及其克隆 被引量:4

Locating and Cloning of Genes Affecting Content of Ginsenosides in Ginseng Hairy Roots

摘要 通过发根农杆菌与人参外植体共培养 ,在人参根外植体上用直接接菌方法 ,诱导出人参发根。Southern点杂交证明 :本研究获得的人参发根确已被Ri质粒的T DNA所转化 ,并被整合到人参发根DNA上。根据Slightom等RiA4TL DNA序列分析结果 ,利用PCR方法从人参发根中扩增并克隆了影响人参皂甙合成基因rolC。 Ginseng hairy roots were induced from the root explants of ginseng by the co culture of Agrobacterium rhizogenes and ginseng roots and the method of direct inoculation.Southern dot blotting proved that the ginseng hairy roots in this study have been indeed transformed by the T DNA of Ri plasmid and the T DNA has been integrated into the DNA of the ginseng hairy roots.Based on the analytical results of RiA 4T L DNA sequence by Slightom et al.,gene rolC affecting the synthesis of ginsenosides was amplified and cloned from the ginseng hairy roots by the PCR method.
