戊二醛和季铵盐复合消毒剂对鸡常见病原微生物杀灭效果的研究

Study on Efficacy of the Mixed Disinfectant of Glutaraldehyde and Quaternary Ammonium Salt against Chicken Pathogen Microorganism

目的 研究戊二醛和季铵盐复合消毒剂对鸡常见的几种病原微生物杀灭效果。方法 将金黄色葡萄球菌、大肠埃希菌和类炭疽杆菌培养 ,用PBS缓冲液和含有有机物的溶液 (5 %鸡粪和 10 %小牛血清 )分别稀释成含菌 5× 10 6CFU ml,分别加入到不同浓度的消毒剂中 ,作用不同的时间之后 ,进行平板细菌计数 ;将含 2× 10 5EID50新城疫病毒 (NDV)和禽流感病毒 (AIV)分别与不同浓度的复合消毒剂作用不同的时间 ,尿囊腔接种 9日龄SPF鸡胚(0 2mL) ,孵育 7d ,测其尿囊液中病毒血凝效果。结果 1∶5 0 0稀释的复合消毒剂在水溶液和有机物溶液中 ,10min能百分之百的杀灭大肠埃希菌和葡萄球菌 ,1∶10 0稀释时 ,在 10min内能杀灭类炭疽芽胞杆菌 ;0 1%的消毒剂在 5min内可有效杀灭NDV和AIV。结论 戊二醛和季铵盐复合消毒剂有很强的杀菌和杀病毒能力 ,即使在有机物存在的条件下 。

Objective To study efficacy of the mixed disinfectant Glutaraldehyde and Quaternary Ammonium Salt against chicken pathogen microorganism.Methods Bacteria including Staphylococcus aureus,Escherichia coli and Bacillus anthracoides Trevisan were diluted into solution with 5×10 6CFU/ml by PBS or organic solution (5% chicken feces or 10% BSA),and then put into various concentrations of the mixed disinfectant of Glutaraldehyde and Quaternary Ammonium Salt for some minutes.Finally,the treated solutions were cultured and germs were counted.On the other hand,the solution containing NDV and AIV with 2×10 5EID 50 were treated by different concentrations of the mixed disinfectant for 5 minutes,after that,the treated solutions were inoculated in SPF chicken embryos with 0.2 mL/embryo,the embryos were hatched for 7 days in 37℃.The viruses were measured by HA test.Result The mixed disinfectant with 1/500 deluted could kill the Staphylococcus aureus,Escherichia coli and the disinfectant with 1/100 deluted could kill the similar of Bacillus anthracoides Trevisan in aqueous or organic solution in 10 minutes.NDV and AIV could be killed by the mixed disinfectant with 10ppm in 5 minutes.Conclusion The mixed disinfectant of Glutaraldehyde and Quaternary Ammonium Salt could kill common bacteria and viruses effectively.The organic solution didn't affect the efficacy against bacteria.
