摘要
用纯化人血栓调节蛋白(TM)免疫Balb/c 小鼠,经细胞融合获得6株抗人TM 单抗,分别命名为SZ—52、SZ—53、SZ—54、SZ—55、SZ—56、和SZ—57。这6株单抗都能与人TM 和内皮细胞结合,并能抑制人TM 活性,而与除TM 外的血浆蛋白无交叉反应。其中SZ—52、SZ—53和SZ—57与红细胞、白细胞和血小板呈阴性反应,为特异性抗人TM 单抗;其余3株单抗则与白细胞或血小板有不同程度的交叉反应。
Balb/c mice were immunized with pruified human thrombomodulin(TM),and thespleen cells of immunized mice were fused with mouse myeloma cells.Six monoclonalantibodies against human TM were obtained,named SZ-52,SZ-53,SZ-54,SZ-55,SZ-56 and SZ-57 respectively.All the six monoclonal antibodies were able to bind tohuman TM and endothelial cells,to inhibit the activity of TM,but had no cross reactionto plasma proteins except TM.Among these,SZ-52 SZ-53 and SZ-57 gave a negativereaction to red cells,white cells and platelets,so they are specific monoclonal antibodiesto human TM,however,the other three monoclonal antibodies had cross reaction towhite cells or platelets.
