^(188)Re作用于三种肿瘤细胞株的剂量-效应关系及敏感性研究

摘要 目的 研究放射性核素1 88Re作用于不同肿瘤细胞株的剂量 效应关系及药物敏感性。方法 采用MTT法观察1 88Re作用于前列腺癌PC 3细胞、乳腺癌ER 75 30细胞和非小细胞肺癌A5 4 9细胞后各细胞株生长受抑制的程度并绘制抑制率曲线。结果 PC 3、ER 75 30、A5 4 9细胞加入1 88Re 48h后 ,细胞形态发生变化 ,表现为形态不规则 ,贴壁能力下降 ,透光度降低 ,部分细胞崩解死亡 ,且效应随着药物浓度的增加更为明显。在同一剂量下抑制率为PC 3>ER 75 30 >A 5 4 9。对1 88Re的敏感性PC 3最敏感 ,ER 75 30细胞次之 ,A5 4 9细胞相对较差。结论 1 88Re对肿瘤细胞的生长具有抑制作用 ,不同肿瘤细胞对1 Objective To study the dose response ralationship and radiosensitivity of three different cancer cell lines after exposure to radionuclide 188 Re. Methods Dose response ralationship and radiosensitivity were evaluated by examining inhibition of radionuclide 188 Re in various concentrations on three cencer cell lines (PC 3, ER-75-30, A549) in vitro with MTT assay.Results After being exposed to 188 Re 48 h, significant dose-response ralationship was shown in the three cancer cell lines. With increasing concentration, inhibition rate rose. At the same concentration, the inhibition rates of three cancer cell lines were different, (PC-3>ER-75-30>A549). They also had different sensitivities to 188 Re. PC 3 cell was the most sensitive to 188 Re and A549 cell was the least sensitive.Conclusion Radionuclide 188 Re can inhibit the growth of cancer cells and different cancer cells have different sensitivity to 188 Re.
