摘要
目的 :利用大鼠泌乳素(rPRL)的纯品和抗体 ,建立rPRL放射免疫分析 (RIA)。方法 :以氯胺 -T法制备125I标记rPRL ,采用平衡竞争法建立rPRLRIA ,并测定大鼠血清和垂体组织中rPRL含量。结果 :本方法灵敏度为0.24μg/L ;批内和批间变异系数分别为2.6 %~8.9%和9.1 %~12.7 %;回收率为90.2%~109.8 %;标准曲线范围为0.39μg/L~50μg/L。用本方法测定了经人心房利钠多肽 (ANP)诱导的大鼠血清及垂体组织中rPRL含量 ,其结果与实验动物实际情况相吻合。结论 :本方法是检测rPRL水平的有效方法 ,为大鼠垂体分泌rPRL的基础研究提供了一种可靠的检测手段。
Objective: To establish the radioimmunoassay(RIA)for rat prolactin(rPRL) using rPRL antigen and anti-rPRL antibody.Methods: Equilibrium competitive rPRL RIA was developed using 125I labelled rPRL,and the method was used to determined rPRL levels in rat sera and pituitary tissues.Results:The assay had a detection limit of 0.24 μg/L. The coefficients of variation within an assay and between assays were 2.6%~8.9% and 9.1%~12.7% respectively. The recovery rate was 90.2%~109.8%. Standard curve range was 0.39 μg/L~50 μg/L.Serum and pituitary tissue levels of rPRL were determined, which were consistent with situation of experimental animals. Conclusion:The method is suitable for use in rPRL level determination.
