摘要
目的 :探讨蕨类植物半边旗提取物 6F杀伤HL 60细胞的作用机制。方法 :应用倒置显微镜观察细胞形态、DNA琼脂糖凝胶电泳、流式细胞光度术、DNA片段形成率等方法检测细胞凋亡。结果 :5 8~ 2 31nmol L 6F作用HL 60细胞 12~ 2 4h后 ,呈现典型的凋亡细胞的形态学改变 ,琼脂糖凝胶电泳可见DNA梯状条带 ,流式细胞光度术结果显示有亚倍体 (Sub G1)峰 ,DNA片段率结果显示 6F诱导HL 60细胞凋亡呈时间和剂量效应关系 ,6F达到 2 9μmol L时只需作用 3h即可直接使细胞坏死。 结论 :6F可诱导HL 60细胞凋亡 ,药物在一定的浓度范围内是其杀伤HL
Objective To investigate the cytotoxicity mechanisms of compound 6F isolated from Pteris Semipinnata L. (PSL).Methods The apoptosis of HL 60 cells has been confirmed by means of light microscopy,DNA agarose gel electrophoresis,flow cytometry and DNA fragment formation test.Results Treated with 58-231 nmol/L 6F for 12-24 hours,HL 60 cells showed typical apoptosis alterations:morphologic changes in light microscopy,DNA ladders on agarose gel,Sub G 1 peak on flow cytometric histogram,and DNA fragment test showed that the induction effect of apoptosis by 6F was in a obvious time and dose dependent manner.Necrosis was shown in HL 60 cells when treated with 29 μmol/L 6F for 3 hours.Conclusion Compound 6F could induce apoptosis in HL 60 cells,and this may explain one of the cytotoxicity mechanisms of 6F within a certain concentration of range.
