摘要
目的 :探讨心肌肌浆网钙调节蛋白功能变化在高血压左心室肥厚病理中的作用。方法 :用自发性高血压大鼠 (SHR)制成左心室肥厚模型 ,以 [3 H]ryanodine为放射配基、p 硝基苯酰磷酸 (PNPP)为底物分别测定左心室肌浆网ryanodine受体密度及钙ATP酶活力的改变。结果 :(1)SHR左心室肥厚时 ,ryanodine受体Bmax值比对照组 (WKY组 )升高 (P <0 .0 1) ,亲和常数 (Kd)无变化 ;钙ATP酶活力下降 (P <0 .0 1)。 (2 )与SHR组相比 ,SHR培哚普利治疗组ryanodine受体Bmax下降 (P <0 .0 5 ) ,钙ATP酶活力上升 (P <0 .0 5 )。结论
Objective The experment was designed to investigate the role of the sarcoplasmic reticulum(SR) Ca 2+ handling protein (ryanodine receptor and Ca 2+ ATPase) in development of left ventricular hypertrophy(LVH).Methods Spontaneously hypertensive rats (SHR) were used as hypertension model and WKY as control,ryanodine was used as a radioligand to assay the function of ryanodine receptor and p nitrophenyl phosphatase was used to analyze the calcium ATPase activity.Results (1)Left ventricular weight/body weight(LVW/BW) in SHR increased significantly compared to that of WKY( P <0.01).No change was found in Kd.Calcium ATPase activity decreased.(2)Compared to SHR group,the LVW/BW in SHR perindopril treatment group decreased ( P <0.05),but the calcium ATPase activity was increased.Conclusion The left ventricular hypertrophy is closely related to AngⅡ mediated the function of SR Ca 2+ handling protein.
