摘要
目的 :探讨重组人转化生长因子 β3 (rhTGFβ3 )在伤口愈合及瘢痕形成中的作用机制。方法 :取体外培养的人正常皮肤和增生性瘢痕成纤维细胞 (normalskinfibroblast,NSFb ;hypertrophicscarfibroblast ,HSFb) ,应用不同浓度rhTGFβ3 ,通过放射免疫和Northernbloting杂交方法观察NSFb和HSFbⅠ、Ⅲ型胶原蛋白及转录水平mRNA表达的变化。结果 :(1)人正常皮肤和瘢痕组织成纤维细胞Ⅰ、Ⅲ型前胶原有明显的不同表达 ;(2 )与对照组相比 ,实验组 (分别加rhTGFβ3 ,终浓度为 1、5、10ng·L-1)Ⅰ、Ⅲ前胶原合成明显增加 (P <0 .0 0 1) ,Ⅰ /Ⅲ前胶原的比例变小 ;(3 )实验组rhTGFβ3 对NSFb和HSFb生物学作用的影响呈明显的量效关系。结论 :rhTGFβ3 可有效提高成纤维细胞Ⅰ、Ⅲ型胶原的合成和Ⅰ、Ⅲ型构成比中Ⅲ型胶原的相对含量 。
Objective To investigate the possible biological role of recombinant human TGF β 3 (rhTGF β 3) on wound healing and scar formation.Methods Using different dose rhTGF β 3 on an in vitro model of cultural human normal skin and hypertrophic scar fibroblasts.The type Ⅰ and type Ⅲ precollagen protein contents in culture medium were determined by radioimmunoassay.The type Ⅰ and type Ⅲ precollagen transcription level mRNA expression were assayed by Northern blotting analysis.Results The results shown that there were different expression of the type Ⅰ and type Ⅲ precollagen in the two kinds of cells.Comparing with control groups (1)The type Ⅰ and type Ⅲ precollagen contents were higher.Moreover,The ratio of the fibroblasts type Ⅰ to type Ⅲ precollagen were reduced.(2)rhTGF β 3 pharmacological action presented a dose dependent manner.Conclusion The exogenous rhTGF β 3 can effectively increase production of the fibroblasts type Ⅰ and type Ⅲ precollagen and their mRNA expression,and relatively increase secretion of target cells for the type Ⅲ precollagen,and may have an important role for accelerate wound healing and reduce scar formation.
