表达鼠精子Cyritestin蛋白的重组沙门氏菌疫苗株的免疫性避孕效应研究 被引量:2

摘要 目的 为观察沙门氏菌疫苗株诱导雌鼠免疫性避孕效应。方法 应用表达鼠精子抗原cyritestin的两种重组沙门氏菌疫苗株X46 32 (pCFL)、X45 5 0 (pCFL)口服免疫雌鼠 ,通过ELISA方法检测其激发雌鼠产生抗重组Cyritestin蛋白和抗菌体LPS(脂多糖 )的血清IgG抗体和阴道分泌物IgA抗体滴度变化。 结果 发现 :两种重组疫苗株不但可激发雌鼠产生高水平血清抗重组Cyritestin蛋白和抗菌体LPSIgG抗体 ,而且在阴道分泌物中可出现相应IgA抗体。同时 ,口服免疫后雌鼠生育能力明显降低 ,两实验组雌鼠妊娠率降低 30 %~ 40 % ,一胎产仔数量平均降低 6 6 .96 %~ 70 .48%。结论 表达精子Cyritestin蛋白的重组沙门氏菌疫苗株可诱导有效的免疫性避孕效应 ,值得深入研究。 Objective To explore the immunological contraceptive effects induced by the X4632(pCFL)?X4550(pCFL),two strains of the recombinant Salmonella typhimurium expressing mouse sperm mature cyritestin protein respectively.Methods The female BALB/c mouse were immunized with X4632(pCFL)?X4550(pCFL),Their serum IgG titres and vaginal secretions IgA titres against to cyritestin protein and Salmonella typhimurium LPS were detected using ELISA assays.Results High level serum IgG titres and some degree vaginal mucosal IgA titres in mice induced by oral immunization were detected in both groups.Meanwhile,the fertility of the two immunized groups was significantly decreased.The pregnant rates of the mice immunized by X4632(pCFL) and X4550(pCFL) were significantly decreased by 30% and 40%,respectively ( P <0.01).Each litter sizes of the two groups were significantly reduced by 66.96% and 70.48%,respectively ( P <0.01).Conclusions Two recombinant strains expressing the mouse sperm mature cyritestin protein can effectively inducing specific humoral and mucosal antibody response and contraceptive effects in Kunming mice.Their charactrization are well worth deep investigation.
