摘要 目的 了解胰岛素样生长因子 - 1(IGF1 )对体外生长的兔关节软骨细胞的生物学效应 ,为组织工程构件软骨提供理论基础 .方法 取第 3代兔关节软骨细胞体外单层培养 ,培养液 DMEM中以终浓度分别为 1,10 ,10 0μg.L- 1 的 IGF1 各作用细胞 2 ,4及 6 d,以 MTT法检测细胞的增殖情况 ,并在 10 0μg.L- 1 IGF1 作用下 ,采用流式细胞技术进行细胞周期亚时相分析 .同时 ,检测基质中糖醛酸的变化反映蛋白多糖含量的变化 .结果 结果显示在 1~ 10 0μg.L- 1 浓度下 IGF1 能明显促进培养软骨细胞的增殖 ,且以 10 0μg.L- 1 作用 4 d刺激效果最明显 ;并能显著影响细胞外基质中糖醛酸的含量 .结论 IGF1 AIM To explore the effects of insulin like growth factor 1 (IGF1) on the metabolism of articular chondrocytes and their matrix. METHODS Monolayer cells of 3 generation obtained from 4 week rabbit were cultured in DMEM containing 10% fetal calf serum and were treated with IGF 1 with doses ranging from 1 to 100 μg·L -1 over 2 d, 4 d and 6 d periods. Effect of IGF 1 was assessed on the proliferation of chondrocytes was measured by MTT method and the glucuronic acidcontent, which reflected the functions of chondrocytes, and on the cyclic change of the cells. RESULTS IGF 1 was found to stimulate cell proliferation and matrix metabolism in a dose and time dependent manner. The maximal effect of stimilation occurred at 100 μg·L -1 within 4 d after treatment. CONCLUSION Cell proliferation and matrix metabolism in the cultured articular chondrocytes can be stimulated by IGF1.
