摘要
目的 观察移植的胚胎心肌细胞在成年兔急性梗死心肌组织内的生长发育过程和其与宿主心肌组织的相互作用 ,以及对周围心肌血运的影响 .方法 将培养的兔胚胎心肌细胞 ,经 DAPI标记后 ,直接注入成年兔的急性梗死心肌组织内 (通过结扎左冠状动脉前降支建立急性心肌梗死模型 ) ,同时设立对照组 .4 wk后处死动物 ,进行组织学和电镜观察 .结果 4 wk后的心肌组织切片中 ,移植心肌细胞与宿主心肌细胞之间形成闰盘 ,其排列方向与宿主心肌肌纤维方向平行 ,同时 ,与对照组相比 ,心肌梗死区面积减少及移植区血管密度增高 (P<0 .0 5 ) .结论 移植的胚胎心肌细胞在受体急性梗死心肌内可以继续生长发育 ,逐渐分化成熟 ,从而稳定病损的心肌结构 ,改善心脏的收缩功能 ,同时 。
AIM To investigate the interaction between implanted fetal cardiomyocytes and host cardiomyocytes and the angiogenesis effects of implanted fetal cardiomyocytes on the host cardiomyocytes and the development of fetal cardiomyocytes implanted into adult acute infarction myocardium. METHODS After labelled with DAPI, cultured rabbit fetal cardiomyocytes were implanted into the actue myocardial in farction site of adult rabbits via direct injection. The myocardial infarction model was established by ligating the left anterior decending artery. Specimens were harvested 4 weeks after the cell implantation. The results were observed by histologic methods and electronic microscopy examination. RESULTS Intercalated disks were observed between the grafted and the host cardiomyocytes, with the implanted cardiomyocytes parallel to the host myocardium in an aligning direction. The area of myocardial infaction decreased and the vascular density of implantationsites increased compared with those of the control groups ( P <0.05). CONCLUSION The fetal cardiomyocytes, after being implanted into the acute myocardial infarction, can differentiate into mature cardiomyocytes in the host heart, which not only repair the structure of the damaged cardiomyocytes and improve the systolic function of the host heart, but also induce angiogenesis in the core of the infarct.
