DDPH和生长因子对肺动脉平滑肌细胞凋亡的作用

摘要 运用原位末端转移酶标记技术 (TUNEL)和流式细胞分析术 (FCM)检测经 1- (2 ,6 -二甲基苯氧基 ) - 2 -(3,4-二甲氧基苯乙胺基 )丙烷盐酸盐 (DDPH)、血小板源性生长因子 (PDGF)、碱性成纤维细胞生长因子 (b- FGF)、低氧内皮细胞条件培养液 (HECCM)作用后 ,肺动脉平滑肌细胞 (PASMC)凋亡的变化 ;结合 FCM对各组 PASMC生长周期的分析 ,比较不同因素对 PASMC的影响差异。结果发现 :DDPH组平滑肌细胞 (SMC)凋亡指数显著高于对照组 ,PDGF、b- FGF、HECCM各组 SMC凋亡指数显著低于对照组 ;PDGF、b- FGF、HECCM3组通过生长限制点 G1 /S(S+G2 /M期 )细胞的比率均显著高于对照组 ,尤以 PDGF组最高 ,而 DDPH组低于对照组。提示 :DDPH能够抑制PASMC增殖并诱导其凋亡 ,而 PDGF、 b- TUNEL and flow cytometry methods were applied to investigate the changes of the apoptotic index (AI) of the cultured pulmonary artery smooth muscle cells (PASMC) after treatment with DDPH, PDGF, b-FGF, HECCM and analyze cell cycle of each group by flow cytometry and the effect of different factors on PASMC was compared. The results showed SMC AI in the DDPH group was significantly higher than in the control groups, while SMC AI in the PDGF, b-FGF and HECCM groups was significantly lower than in the control groups. The cell ratio in S+G 2+M phase in the PDGF, b-FGF, HECCM groups were higher than in the control groups, especially PDGF group with the highest, while that in the DDPH group was lower than in the control groups. The results indicated that DDPH could suppress PASMC's proliferation and induce their apoptosis, but the effect of growth factors such as PDGF and b-FGF was contrary.
